If you are planning to remove this door and permanently close the opening, the door and the door frame must be removed properly. The opening will then be covered with a timber frame, insulation installed and then plasterboard placed on top. The gaps will then need to be filled with filler and the surface primed and painted.

Thank you for your insight Eric! Will this work with a concrete wall as well? The stud wall would need to either be drilled into the concrete sides or do you recommend using liquid nail? Also, since this doorway goes to the backyard, it would need rendering to match the outside of the house, would this be okay? Is blue board not an option for something like this?

Yes, if you are talking about the James Hardie HardieTex™ 2440 x 1200 x 7.5mm Cladding 2.93m². It can be used for cladding, and you can render over it. The timber stud would need to be drilled into the concrete in combination with liquid nails. Please remember to install a moisture barrier and insulation into the timber frame. However, because this is an exterior door, I suggest double checking with a registered builder in regards to its removal.
I also suggest checking the rules and regulations of your local council in regards to exterior door removal. A permit might be required, and a licensed trade might be needed to accomplish the installation.
