My plan is to cut a piece of cladding to fit the hole and screw it to the timber beam, then fill the gaps and cover with render, but I need to ensure it is waterproof, as it is an exterior wall. What type of cladding and filler should I use?

Hardieflex would be an appropriate cement sheeting to use for the repair. Parfix 500g Ready To Use Multipurpose Filler can then be used to fill gaps. You'll finish off by using render to cover the repair and blend in with the surrounding wall. Here's a helpful guide: How to repair a rendered wall.
The key to starting this repair will be adequately mounting your cement rapir sheet. I suspect you'll need to find more mounting points than just the exposed timber. You can use Liquid Nails to adhere small timber blocks to the back of the damaged sheet around the edge. Once the glue is dry, you can adhere your repair sheet to them.
